I am an electrical engineer launching an RFID business. Are inlay manufacturers willing to provide message definitions sent out by the interrogator, so that I can use my own reader for testing?

The commands sent by an RFID reader are determined by the air-interface protocol standard, so if you are using any standard tag—low-frequency (LF), high-frequency (HF) or ultrahigh-frequency (UHF)—you should be able to download the specifications for the standard and utilize your own interrogator to communicate with tags.
